    Product Background Overview

     

    A CAR is an engineered receptor designed to modulate and redirect immune cell responses toward tumor-associated antigens of intrinsic origin. This receptor is a chimeric protein composed of four principal components: an extracellular antigen-binding domain (e.g., a single-chain variable fragment or scFv), a spacer or hinge domain, a transmembrane domain, and at least one intracellular signaling domain. The successful integration of these components enables CAR-expressing immune cells to recognize specific antigens and kill tumor cells.

    During the discovery and preclinical research phases, when engineering CAR-X cells, scientists must verify cellular functionality by monitoring the surface expression of the CAR. Most existing detection methods rely on reagents that bind to the extracellular domain of the CAR and exploit specific sequences within the scFv, the variable heavy-chain domain of the hinge region, or the variable light-chain domain. Depending on the class of CAR-X detection reagent used, the available solutions often lack sufficient specificity, are difficult to integrate into multiplex flow cytometry panels, or are limited to detecting only a single CAR.

    Currently, common methods for detecting CAR expression include using Protein L, target antigens, tag-specific antibodies, anti-idiotype antibodies (ADA), and co-expression of GFP or tEGFR; however, each of these approaches has its own advantages and limitations, and some are not suitable for practical applications.

    In 2010, two patients with hematologic malignancies became the first participants in a clinical trial of CAR-T cell therapy at the Perelman School of Medicine at the University of Pennsylvania, and their cancers achieved complete remission that same year. More than a decade later, research published in Nature and based on up to 10 years of follow-up data show that CAR-T cells can still be detected in patients’ bodies 10 years after infusion. Owing to their high selectivity, localized high concentrations, and customizable nature, cell therapies have ushered in a new chapter in cancer immunotherapy.

    The manufacturing process for CAR-X cell therapies is complex and involves numerous quality-control steps. As an essential component of quality assurance for CAR-X therapy, the assay for detecting the positive expression rate of CAR-X currently lacks a standardized protocol. The Anti-(G4S)n (B02H1) monoclonal antibody, a broadly applicable CAR detection reagent, can directly identify and confirm the presence of CAR regardless of its antigen specificity. By recognizing the flexible (G4S)n linker, this antibody can bind to CARs with diverse specificities expressed on the cell surface; such CARs all contain a (G4S)n spacer (n ≥ 2) in their extracellular scFv domain.

    Based on clinical needs and industry pain points, the HeYouSheng Bio team launched this unique product—Anti-(G4S)n linker (B02H1) mAb—domestically for the first time four months ago.     Anti-GSlinker (B02H1) It has the following characteristics:

    4. High sensitivity: It exhibits excellent detection efficiency at both extremely high and extremely low concentrations.
