    Using the HeYouSheng NK serum-free culture system (cytokine-based method), PBMCs from batch 059K025 were cryopreserved at an initial cell count of 100 million. On Day 7, aliquots of 5 × 10^7, 1 × 10^8, and 2 × 10^8 cells were seeded into Cell Feeder底透 cell culture flasks, with the medium volume directly supplemented to 1 liter. The remaining 1 × 10^8 cells were cultured in bioreactor bags, with density-adjusted medium replenishment performed as usual.

    Steps for Using Cell Feeder Bottom-Permeable Cell Culture Flasks

     

    Cell Feeder bottom-breathable cell culture flasks are suitable for the expansion and cultivation of various non-adherent cells. This product employs a dual-layer breathable culture system: during the culture process, cells settle on the bottom breathable membrane, allowing them to efficiently absorb nutrients from the culture medium while also receiving ample oxygen through the membrane, thereby promoting rapid cell proliferation.

     

    Usage steps:

    1. The initial activation and expansion are conducted exactly as in bag culture and must be performed in T-flasks.

    2. After the cells have been thoroughly resuspended in the culture flask, first add 100 mL of culture medium and gently shake the flask to ensure uniform distribution of the cells across the bottom; then immediately add 900 mL of culture medium (including all supplements required during the culture process).

    3. Transfer the bottom-permeable bottles into the incubator and allow them to incubate undisturbed for 7 days; after 7 days, remove them and harvest the culture.

    4. During harvest, it is recommended to use a pipette or peristaltic pump to remove 80–90% of the culture medium. The pipette tip should be moved slowly from top to bottom to the desired cell layer height, avoiding direct insertion into the bottom to prevent excessive suction that could dislodge cell clumps. After removing 80–90% of the medium, gently shake the flask to resuspend the remaining cells and medium, then tilt the flask slightly and use the pipette to aspirate the remaining cells and medium.

    Caution: When aspirating, do not allow the pipette tip to come into direct contact with the bottom membrane of the透瓶.

    Q5: Approximately what total cell count can be achieved when culturing cells in a cell-permeable culture flask?

    Answer: The cell count is influenced by factors such as the cell type being cultured, the composition of the culture medium, the duration of cultivation, and the initial cell seeding density, among others; results vary depending on these conditions. Typically, a cell density of 1 million to 10 million cells per milliliter is observed.

     

    Q6: Is cell counting required during the culture process?

    Answer: We do not recommend performing daily cell counts. Data indicate that frequent counting during the culture process can alter the intracellular microenvironment, resulting in a 10–20% reduction in yield. Additionally, as cells tend to settle at the bottom of the culture vessel during cultivation, sampling either the supernatant or the cell pellet at the bottom can lead to inaccurate technical data.

     

    Q7: Can culture flasks be reused?

    Answer: No, bottom-breath culture flasks are not for re-sterilization and must be used only once.
