Introduction to the Anti-FMC63 Antibody

 

CD19 is currently the most widely used target in CAR-T cell therapy, and numerous studies have demonstrated that CD19 is an effective and safe target for the treatment of B-cell acute lymphoblastic leukemia (B-ALL), chronic lymphocytic leukemia (CLL), and B-cell lymphomas. FMC63 is a murine monoclonal antibody that targets human CD19. To date, the majority of reported CAR-T clinical trials have constructed CAR-T cells based on the FMC63 antibody sequence, including the two FDA-approved anti-CD19 CAR-T products, Kymriah and Yescarta. According to the “Research Guidelines on Quality Control Testing and Nonclinical Considerations for CAR-T Cell Therapy Products” issued by the National Institute for Food and Drug Control in 2018, flow cytometry should be used to assess CAR transduction positivity, with preference given to antigen-specific proteins or anti-idiotypic antibodies that exhibit superior specificity for the antigen-binding site.

 

Following the launch of a pan-specific G4S-positive-rate detection antibody, HeHycells Biotechnology has now introduced a unique-epitope–targeting anti-idiotypic antibody that specifically recognizes the FMC63 scFv antigenic epitope, thereby further expanding the array of assays for assessing CAR positivity. This antibody is a rabbit monoclonal antibody characterized by high affinity, high sensitivity, and excellent specificity, and has been thoroughly validated for use in flow cytometry-based detection of Anti-CD19 (FMC63) CAR expression.

Experimental Materials and Validation Data

 

 

 

Cell:

293T-CAR19 stable transfectant cells

 

Primary antibody:

FMC-63-PE (HY-D1857F) and FMC63-APC (HY-D1858F) were added to 293T-CAR19 cells in EP tubes at a final concentration of 1 µg/mL, followed by incubation in the dark at room temperature for 30 minutes.

 

Secondary antibody:

Add DyLight™ 649 donkey anti-rabbit IgG (minimal cross-reactivity) antibody to the EP tube at a 1:250 dilution, and incubate in the dark at room temperature for 30 minutes.

 

Protein:

APC-labeled monoclonal anti-FMC63 scFv antibody and PE-labeled human CD19 (20–291) protein were each added to 293T-CAR19 cells in EP tubes at a 1:50 dilution, followed by incubation at room temperature for 30 minutes.
