China’s First Launch—New Tool for Detecting CAR Positive Rate
In 2010, two patients with hematologic malignancies became the first participants in a clinical trial of CAR-T cell therapy at the Perelman School of Medicine at the University of Pennsylvania, and their cancers achieved complete remission that same year. More than a decade later, research published in Nature and based on up to 10 years of follow-up data indicate that CAR-T cells can still be detected in patients’ bodies 10 years after infusion. Owing to their high selectivity, localized high concentrations, and customizable nature, cell therapies have ushered in a new chapter in cancer immunotherapy.

The manufacturing process for CAR-X cell therapies is complex and involves numerous quality-control steps. As an essential component of quality assurance for CAR-X therapy, the assay for detecting the positive expression rate of CAR-X currently lacks a standardized protocol. The Anti-(G4S)n (B02H1) monoclonal antibody, a broadly applicable CAR detection reagent, can directly identify and confirm the presence of CAR regardless of its antigen specificity. By recognizing the flexible (G4S)n linker, this antibody can bind to CARs with diverse specificities expressed on the cell surface; these CARs all contain a (G4S)n spacer (n ≥ 2) in their extracellular scFv domain.
